I can't seem to get the tube out. Is there a set screw or equivalent I need to deal with ? There does seem to be a machined hole on the front of the motor in that location but it is real hard to get at wqithout removing a number of brackets etc. I thought this would be quick and easy.

There was no set screw or other retaining device ever ORIGINALLY used for the oil fill tube. However, that doesn't mean that someone "along the way" might not have added one to "compensate" for the loss of the original interference fit. I think that it's VERY unlikely, though.
The best way to remove these tubes is with the manifold off the car. Then, they can be driven out from the underside. This is especially true for "stubborn" cases (like, apparently, yours). If these tubes have been in place for a long time (like, since the engine was built) they will tend to "weld" themselves to the manifold.
