69 Convertible Corner windshield moldings need replacing. They appear to be secured with one screw and some type on silicone or clear adhesive. What should I use to remove these, and what should I secure the moldings with?

Andy, no clear or silicone adhesives were used anywhere on early C3 Corvettes. You have a "Bubinstallation" there.
If you are referring to the cast, chrome-plated part with the nylon-bushed hole for the convertible top guide pin, this part should actually have three chrome plated, crown-headed, countersinking screws (two on the passenger side of the header and one on the outside end of the part). You may also find that you are missing the sheet metal "J-nut" clips on the bird cage header into which the metal screws thread.
With three screws, adhesive serves no purpose, but originally there was a pliable gray sealing compound that filled the void under this part...today's best equivalent would be 3M "Strip-Kalk" available at auto paint stores. I think it may only come in black, but color is immaterial since this sealant can't be seen anyway.
It sounds like "Bubba" used silicone sealer. Take a utility knife and gently cut the silicone. This will reduce the chance of damaging the molding when you remove it.
