You would start with the 396 only features such as the battery on the driver's side (same as AC cars). The serial number would be another clue as they were introduced mid year (not sure what # they started). I am sure there are other items but if they wanted to "make" it a big block it could be done. Hi;
A couple of other things to look for... Check the rear end, the BB rears had special codes, also check the date on the rear, it should jive with the build date etc. It should also have the BB half shaft caps versus the "U" bolts found on SB cars. On the block, (assuming the "original" engine) check for the two gussets just under the intake... later blocks did not have them even though they had the same part number. In the rear, the sway bar assembly.
The car should have all of these (including the battery on the left and the removable access panel for the battery AND no evidence of AC). It is possible (and maybe probably) that the original block is long gone... in that case stick with the other items.
I went through this process myself on my '65 and depending upon what you have and don't have, putting one of these back into reasonable shape can cost $$$$.

One other thing, which I found to be true on my case.... Everything under the hood; wire harnesses, hoses, etc., were dried out and fairly brittle... probably owing to the greater heat generated by the Big Block!!!- Top

The big and easy give away most the guys forget when turning a 66 into a BB is the fuel line. If the body has never been off (check rear area bolts for wrench socket marks), the fuel line is an easy give away.

There was a fuel line photo somewhere here also. BB makes a "sharper turn" from the frame (top) towards the fuel pump. Like 90 degrees.https://MichiganNCRS.org

The end is actually forward of the motor mount frame bracket so what happens is the SB fuel line has to be bent back at a funny angle to then swing and reach the forward position of the 427 fuel pump.- Top

You're hitting on ONE of the 'acid' tests! Those early 396 cars (Feb & Mar) build are rather rare. YES, there were BB's built prior to Feb, but those which have been documented were some form of engineering test 'mule' and not true production vehicles for general public sale...- Top
